Yes the '65 TF big block trans is more desirable in the sense that it is cable operated and has the slip yolk. 

Yes, as Mark notes, the Imperials used a transmission set up for two piece driveshaft. But I assume you could swap out that Imperial tailshaft for one that works with a single driveshaft.
