I tried this and found that with the pump configuration the mechanical pump would work alone until about 1/2 tank then I would starve for fuel and had to turn on the electric pump. Pumping through a mechanical pump is dangerous. If there is a diaphram failure in the mechanical pump you can fill the crankcase with fuel which can result in an explosion and fire. If you are running an electric fuel pump use a safety device like a low oil pressure switch to turn off the fuel pump when the engine is not running. The worst case is an accident. I use one of the commercial oil pressure switches designed for the fuel pumps however I have a center off switch that I can run through the oil pressure switch or bypass it to prime the carb. There is also an electronic unit that works similar to new cars. When the key is turned on the electric pump runs for a few seconds to prime the system. Then it takes a coil signal to let it know the engine is running and enables the fuel pump. Just my thoughts, please correct me if I am wrong. the best depends on what your plans are, race, street, etc. you can remove , and use a plate to block the pump/block mount, but you may want to run both. The regulator depends on the pump used. Some are regulated internally, at around 5 or 7 lbs, same aren't. If the pump you choose has a running pressure of 11, or 12 lbs, you will need a regulator to keep pressure down at the carb.
