You definitely have a plug wire or wires wrong. The only way you can get a back fire thru the carb is if the plug fires while the intake valve is open. The only way that can happen is if the plug wire is in the wrong position in the cap. It is very easy to be off one wire.  Been there done that! Once you are sure of the firing order & get it to run then you can address the mixture settings,
