I have thought about it. I am familiar with  EFi and taught the subject for 7 yeares at provincial trade school. 
First thing to remember is the efi doesn\t know or care what type of motor is under it so you can fit any system if you have it complete with ease. 
Watched a former student make a double  throttle body  top for a mustang efi manifold on his 302 and then stuff the whole deal into a Capri. It worked good enough he had some one come along and buy it from him. 
He made the manifold top from 4x4 box tubing for the plenum and steel tubing of the proper size underneath.  
For a big block Mopar my gut instinct would send me looking for a 460 Ford  truck system . I know it would handle the airflow and fuel delivery and would be an excellant jumping of point . After that an aftermarket computer or power chip wpuld probably do the job. 
Also have a German Hot rod magazine with an article on how to convert an early hemi Hilborn unit to Gm off the shelf injectors and a britsch made computer.  I am not sure where it is right now but I know I did not throw it out. . Bought it in the Worms Deutschland train statrion as a Present to myself on my 50th birthday.
However, I thin k Cam design is an area we need to look into as well. 
We have with the mopar cams the ability to open the valves at a tremedous rate due to the .904 lifter diameter. Add to that a set of 1.6 rocker or even the 1.75s Erson used to make (and hopefully still do) for the big block mopar power we could run a cam with high velocity action with a duration at .050 of say 231 and yet have the duration at 200" (or even .100") meet or exceed our current big  pro street cams. I know it would need a little tweaking but I want to play with it to see what happens. 

I think if I become rich I would really like to build a lightened PT Cruiser  wih only 2 doors (Panel van) and a new 5.7Hemi done to the nines. However It is not likely to happen.
