If all systems are in good shape as you have outlined, car probably just
needs good long LOPE down the road.

I would suggest that the engine has not reached proper operating temperature
with your short trip starting.

The choke is intended to richen up he mixture to aid cold start but if the
mechanism is set too rich or the pull off is broken, you will foul plugs.
Intake manifold heat riser passage plugged? Plugged or blocked off passage
will cause exactly the problem you have outlined. Heat riser valve in
exhaust manifold is critical part of cold start system and drives exhaust
through heat riser passage under carburetor when engine is cold to help
vaporize fuel.  Put aftermarket duals on a vehicle and eliminate the riser
valve and the whole cold start system has just been destroyed.

Power to the ignition coil must be correct so that the (in good condition)
coil can produce proper power. Spark output BTW must be varified at the end
of the plug wires. Plugs will foul without proper firing voltage.

I would suggest getting the engine started and let it run at a high idle for
a good period of time (with you there to observe) until at least the
thermostat opens. . High idle would be say 1800 to 2000 RPM. Choke must be
off.

If rough running persists after a few minutes, prepare the power brake
booster vacuum feed line before starting so that you can instigate one big
leaning out vacuum leak. A little running with the line disconnected on-off
will normally clear an overrich condition, clean the plugs up a bit and get
some of the goo off the valve seats, pretty fast. Constant engine speed is
suggested as these engines like to backfire through the carburetor so
goosing the throttle could get a fire started (need 3-A:40-B-C dry chemical
extinguisher on hand, right?).
